Authorities have not released any further details about the victims.
Emergency services were called about the blaze- which had broken out in the kitchen- at around 10pm on Saturday.
The female resident of the detached property sustained third-degree burns.
The Orihuela Policia Local were first on the scene and saw that a neighbour was performing CPR on the 77-year-old.
Officers used a defibrillator before an ambulance team took over for 45 minutes.
There was no response and after around 45 minutes, medics confirmed her death as a result of a suspected cardiac arrest.
Two women- of undisclosed ages- and a 78-year-old man were given initial treatment for smoke inhalation before being taken to Torrevieja Hospital.
The fire itself was quickly extinguished with the main damage in the kitchen area, though the exact cause has so far not been revealed.
Operations at the home ended at 11.45pm.
